It depends: I listen to the whole range and try to understand if there is a pattern in mis-tuning: maybe the last octave is out of tune, or so.
Or, I just have a few notes down, so I just tune those notes. I don't care of the overall pitch, for example the whole harpsichord has gone down or up without severely ruining the temperament. In this case I just tune the few out-of-tunes notes.
Only when I have been away for a while and I find the entire harpsichord put of tune without any recognizable pattern, I decide to tune from scratch. In this case I use my electronic device to get the C in the desired temperament, setting the overall pitch to A=415.3.
But I start from C, and tune by ear, so I guess my A could be lower or higher than 415.3.
